Anti Aging Science1-Methylhydantoin-2-imide (Tego Cosmo). Blocks the enzyme that produces melanin to reduce discoloration.

Ascorbic Acid Polypeptide. A reaction product of ascorbic acid and hydrolyzed protein.

Alpha Hydroxy Acids.  Anti aging skin care ingredients that aid exfoliation by weakening the links between cells in the outer layer of dead skin, allowing the normal shedding process to occur at a more optimum rate.  By helping new cells surface, they improve skin texture, unclog pores and facilitate absorption of moisturizers.  It may also help play a role in building skin-firming collagen and elastin, especially when incorporated into a two-step chemical peel.  AHAs include lactic (from dairy products), glycolic (from sugar), malic (from apples) and citric (from citrus fruits).  Dr. Gross has found that skincare products with several different kinds of acids (AHA and BHA) are the most effective and least irritating, because when many acids are combined, each can be used in a lower (i.e. milder) concentration.

The bad news: As we age, our bodies no longer produce sufficient antioxidants to fight free radicals. The good news: Just as there are different species of free radicals, modern science has identified different kinds of antioxidants to combat them.  Dr. Gross recommends using products containing as many different kinds of antioxidants as possible.

Ayurvedic Medicine.  Refers to a form of holistic, alternative medicine developed in India.  The term “ayurveda” derives from two Sanskrit words: ayu, meaning life, and veda, meaning science.  Ayurvedic skincare focuses on plant extracts and essential oils that produce some of the most effective anti aging skin care ingredients.

Beta Hydroxy Acid.  Also known as Salicylic Acid. Emollient. An ingredient that softens the skin.  Similar in structure to natural lipids found in the skin, helps prevent dryness, soothes irritated skin and makes rough skin smooth.

Enzymes. Enzymes are special proteins that exist in the cells of all living beings. Their purpose is to facilitate naturally occurring biochemical reactions.

Natural Anti AgingEpilobium Angustifolium Extract (Willowherb Extract). A safe, natural anti-inflammatory that works quickly to sooth irritated skin.

Essential Fatty Acids. Sometimes referred to as Vitamin F, essential fatty acids are long-chain polyunsaturated fatty acids derived from linolenic, linoleic  and oleic acids and are crucial to our body’s basic functioning. Since the body cannot make them they must be supplied by food or supplements, which is why they are called “essential” nutrients.  Not only do they regulate several of our bodily functions, including the inflammatory response to injury, they also form part of the structure of cell membranes and are essential for rebuilding cells and producing new ones.  Studies show that EFA’s are essential to the normal health, function, and beauty of the skin.  They can be found in vegetable oils (grape seed, evening primrose, sesame and soybean) and in seeds, nuts, legumes and whole grain products.

Exfoliant.  An ingredient or device, such as a washcloth or loofah, that revs up cell turnover by sloughing off dead surface cells.  Improves skin texture, evens tone and increases radiance.  Exfoliating acids such as Alpha Hydroxy and Beta Hydroxy Acids may also play a role in building skin-firming collagen and elastin.  In peels, it is not only the exfoliating action of acids that increases collagen formation, but the pH fluctuation the skin experiences as it goes from acidic to neutral.  Dr. Gross has coined the term Phlux to refer to this powerful action, which appears to be an even bigger catalyst in cell renewal.

Anti Aging ScienceFarnesol.   Widely found throughout the plant world and in many essential oils. It inhibits bacterial growth and has antioxidant properties.

Fibroblasts.  Connective-tissue cells that secrete proteins, especially collagen, from which the extracellular matrix of connective tissue forms.

Free Radicals.  Chemicals that destroy collagen and lead to wrinkles, lines, etc.  New research shows that there are at least two different kinds of free radicals – those triggered by the body (metabolic) and those sparked by the sun and other environmental aggressors, such as pollution and smoke.  Both types function like little darts, destroying skin-supporting collagen and elastin, but respond to different antioxidants.  This is why skincare products that incorporate as many different kinds of antioxidants as possible are the most effective.

Pea Extract. See Pisum Sativum.

Pentapeptide-4. A synthetic peptide containing lysine, serine and threonine.

A new study demonstrates what researchers consider conclusive evidence that the red wine compound resveratrol directly activates a protein that promotes health and longevity in animal models. What’s more, the researchers have uncovered the molecular mechanism for this interaction, and show that a class of more potent (anti aging) drugs currently in clinical trials act in a similar fashion. Pharmaceutical compounds similar to resveratrol may potentially be used as anti aging treament and prevent diseases related to aging in people, the authors contend.

These findings are published in the March 8 issue of Science.

For the last decade, the science of aging has increasingly focused on sirtuins, a group of genes that are believed to protect many organisms, including mammals, against diseases of aging. Mounting evidence has demonstrated that resveratrol, a compound found in the skin of grapes as well as in peanuts and berries, increases the activity of a specific sirtuin,SIRT1, that protects the body from diseases by revving up the mitochondria, a kind of cellular battery that slowly runs down as we age. By recharging the batteries, SIRT1 can have profound effects on health.

“In the history of pharmaceuticals, there has never been a drug that binds to a protein to make it run faster in the way that resveratrol activates SIRT1,” said David Sinclair, Harvard Medical School professor of genetics and senior author on the paper. “Almost all anti aging drugs either slow or block them.”

In 2006, Sinclair’s group published a study showing that resveratrol could extend the lifespan of mice, and the company Sirtris Pharmaceuticals, which was started by HMS researchers, was founded to make drugs more potent than resveratrol. (Sinclair is a co-founder of Sirtris, a GlaxoSmithKline company, and remains a scientific advisor. Sirtris currently has a number of sirtuin-activating compounds in clinical trials.)

But while numerous studies, from Sinclair’s lab and elsewhere, underscored a direct causal link between resveratrol and SIRT1, some scientists claimed the studies were flawed.

The contention lay in the way SIRT1 was studied in vitro, using a specific chemical group attached to the targets of SIRT1 that fluoresces more brightly as SIRT1 activity increases. This chemical group, however, is synthetic and does not exist in cells or in nature, and without it the experiments did not work. As a response to this, a paper published in 2010 surmised that resveratrol’s activation of SIRT1 was an experimental artifact, one that existed in the lab, but not in an actual animal. SIRT1 activity in mice was, the paper claimed, at best an indirect result of resveratrol, and perhaps even a sheer coincidence.

As a result, a debate erupted over the particular pathway that resveratrol and similar compounds affected. Does resveratrol directly activate SIRT1 or is the effect indirect? “We had six years of work telling us that this was most definitely not an artifact,” said Sinclair. “Still, we needed to figure out precisely how resveratrol works. The answer was extremely elegant.”

Sinclair and Basil Hubbard, then a doctoral student in the lab, teamed up with a group of researchers from both the National Institutes of Health and Sirtris Pharmaceuticals to address this question.

Anti Aging Tablets MedicineFirst, the team addressed the problem of the fluorescent chemical group. Why was it required for resveratrol to rev up SIRT1 in the test tube? Instead of dismissing the result as an artifact, the researchers surmised that the chemical might be mimicking molecules found naturally in the cell. These turned out to be a specific class of amino acid, the building blocks of proteins. In nature, there are three amino acids that resemble the fluorescent chemical group, one of which is tryptophan, a molecule abundant in turkey and notable for inducing drowsiness. When researchers repeated the experiment, swapping the fluorescing chemical group on the substrate with a tryptophan residue, resveratrol and similar molecules were once again able to activate SIRT1.

“We discovered a signature for activation that is in fact found in the cell and doesn’t require these other synthetic groups,” said Hubbard, first author of the study. “This was a critical result, which allowed us to bridge the gap between our biochemical and physiological findings.

“Next, we needed to identify precisely how resveratrol presses on SIRT1′s accelerator,” said Sinclair. The team tested approximately 2,000 mutants of the SIRT1 gene, eventually identifying one mutant that completely blocked resveratrol’s effect. The particular mutation resulted in the substitution of a single amino acid residue, out of the 747 that make up SIRT1. The researchers also tested hundreds of other molecules from the Sirtris library, many of which are far more powerful than resveratrol, against this mutant SIRT1. All failed to activate it.

The authors propose a model for how resveratrol works: When the molecule binds, a hinge flips, and SIRT1 becomes hyperactive.

Although these experiments occurred in a test tube, once the researchers identified the precise location of the accelerator pedal on SIRT1 — and how to break it — they could test their ideas in a cell. They replaced the normal SIRT1 gene in muscle and skin cells with the accelerator-dead mutant. Now they could test precisely whether resveratrol and the drugs in development work by tweaking SIRT1 (in which case they would not work) or one of the thousands of other proteins in a cell (in which they would work). While resveratrol and the drugs tested revved up mitochondria in normal cells (an effect caused activating by SIRT1), the mutant cells were completely immune.

“This was the killer experiment,” said Sinclair. “There is no rational alternative explanation other than resveratrol directly activates SIRT1 in cells. Now that we know the exact location on SIRT1 where and how resveratrol works, we can engineer even better molecules that more precisely and effectively trigger the effects of resveratrol.”

The researchers plan on continuing academic-industry collaborations with the goal of bringing to fruition anti aging drugs that treat diseases associated with aging.

And it’s true for both hard and soft contact lenses, according to this new study involving identical twins

While contact lenses might improve your eyesight without the need for clunky glasses, they might also be having an unintended effect on your appearance. According to a new study published in Aesthetic Surgery Journal, use of contact lenses seemingly leads to eyelid droopiness.

Researchers reviewed photographs of 96 sets of identical twins who met in Twinsburg, Ohio, on a yearly basis from 2008 to 2010, measuring the level of eyelid droopiness (called “ptosis”) in each.

These 39-year-old female identical twins had differing levels of ptosis. The patients were visually identified from a twins database and had individual ptosis measured in each eye. The twin on the left did not wear contact lenses; the twin on the right did wear contact lenses.

Nine different environmental factors were considered as a potential cause for ptosis, and they were evaluated through an extensive questionnaire and special standardized photography to determine the degree of eyelid droopiness in each twin.

According to the data collected, wearing either hard or soft contact lenses was associated with ptosis. The average difference in eyelid droopiness between twins was 0.5 millimetres. Among twins who didn’t wear contacts, ptosis was around 1.0 millimetre. In twins who wore soft contacts, that number increased to 1.41 millimetres, rising further to 1.84 millimetres for those who sported hard contacts. (Luckily, with the advent of soft lenses, hard lenses have all but disappeared.)

These 65-year-old female identical twins had differing levels of ptosis related to wearing contact lenses. The subject on the left wore soft contact lenses and had less ptosis than her twin; the subject on the right wore hard contact lenses and had more severe ptosis than her twin.

The researchers also looked for links between ptosis and BMI, smoking, sun exposure, alcohol consumption, work-related stress, and sleep — none of which had a statistically significant impact on eyelid droopiness. The association between contact use and ptosis has been long assumed but never proven, says lead study author Bahman Guyuron, MD, FACS, a facial plastic surgeon based in Cleveland, Ohio.

“Since the identical twins are genetically destined to have similar facial and eyelid features, if there is a difference, it is primarily related to the environmental factors,” Guyuron tells Yahoo Health. “We were able to demonstrate that, of the external factors unrelated to the genes or ageing, use of contacts was the only factor that linked to the droopy eyelids.”

Guyuron says the effect they saw was “related to the weakening of the muscles that lift the upper eyelid, and not related to loose or redundant eyelid skin.” And while the mere-fractions-of-a-millimetre difference might seem small, it’s enough to affect vision and appearance.

“Even a minimal droopiness of the eyelids denotes lack of vigor, tiredness, and getting old — often prematurely on the contact lens users,” says Guyuron. And in a culture constantly seeking the fountain of youth, this may matter to some.

Guyuron says that plastic surgeons regularly perform a couple of key procedures for droopiness. According to data from the American Society for Aesthetic Plastic Surgery, 165,714 eyelid procedures and 31,315 brow lifts were performed on patients in 2014.

“Correction of droopy eyelids is one of the simplest and most rewarding surgeries that plastic surgeons do,” says Guyuron. “It is usually a very short procedure, it often requires very minimal recovery, and it makes the patients look younger and more energetic.”

Of course, eyelid droopiness will occur with each passing year anyway — and you can always switch to glasses if you’re concerned about the effects of contact use.
